Später beginnt sie für den britischen Geheimdienst zu arbeiten, fast immer unterstützt von ihrem treuen Gefährten Willie Garvin, und erlebt zahlreiche oft höchst gefährliche Abenteuer. Neben Romanen und Filmen hat Modesty Blaise ihre erfolgreichsten Auftritte im Comic. Am 13. Mai 1963 erschien in der englischen Tageszeitung The Evening Standard der erste Comicstrip mit Modesty Blaise. Niemand konnte damals ahnen, dass diese Reihe für mehrere Jahrzehnte überaus erfolgreich, mit mehr als 10.000 Strips, veröffentlicht werden würde. Fünf Zeichner illustrierten ihre Abenteuer bis 2001, angefangen mit Jim Holdaway (1963 - 1970), gefolgt von Enrique Badía Romero (1970 - 1978), John M. Burns (1978 - 1979), Patrick Wright (1979 - 1980), und Neville Colvin (1980 - 1986). Danach kehrt Enrique Badía Romero als Zeichner zurück und begleitet die Serie bis zum letzten Strip, der am 11. April 2001 erschienen ist.
