Background
Cyclin A2 is a pivotal regulatory protein that orchestrates S-phase progression and the G2/M transition by binding and activating cyclin-dependent kinases Cdk2 and Cdk1, thereby phosphorylating substrates that initiate DNA replication origin firing, centrosome maturation, and chromosomal condensation; its expression rises during late G1, peaks in S and G2, and is abruptly degraded via APC/C-mediated ubiquitination in prometaphase to prevent premature mitotic exit, and deregulation—through gene amplification, transcriptional upregulation, or impaired destruction—drives genomic instability and oncogenesis, making it both a biomarker and therapeutic target in multiple cancers.
