Background
Transgelin 2, also known as TAGLN2 or SM22β, is a 22-kDa actin-binding protein belonging to the calponin family, widely expressed in both smooth muscle and non-muscle cells, where it stabilizes and remodels the actin cytoskeleton by cross-linking and bundling F-actin, thereby regulating cell shape, motility, immune synapse formation, and vesicle trafficking; notably, it is markedly up-regulated during dendritic-cell maturation and T-cell activation, promotes tumor growth, invasion, and drug resistance in many cancers, yet exogenous cell-permeable transgelin-2 peptides can enhance dendritic-cell homing to lymph nodes and potentiate anti-tumor immunity, highlighting its context-dependent roles as both an oncogenic factor and a potential immunotherapeutic agent.
