Background

CD352 (also known as SLAMF6, NTB-A, or LY108) is a type I transmembrane glycoprotein belonging to the signaling lymphocytic activation molecule (SLAM) family and a member of the CD2 subfamily of immunoregulatory receptors. This protein is expressed on the surface of natural killer (NK) cells, T cells (upregulated upon activation), and B cells, functioning through homophilic interactions to regulate NK cell cytotoxicity, T cell co-stimulation (independent of CD28), Th1 cytokine production, and B cell tolerance. The extracellular domain of CD352 contains Ig-like domains, while its intracellular region harbors conserved tyrosine-based signaling motifs that can be phosphorylated and recruit adaptor proteins such as SH2D1A (SAP) and SH2D1B (EAT-2), thereby modulating innate and adaptive immune responses. Studies indicate that reduced CD352 expression in multiple myeloma (MM) may be linked to its role in immune regulation and the tumor microenvironment. Additionally, CD352 activation promotes T cell proliferation and cytokine secretion while triggering cytolytic activity and signal transduction cascades in NK cells.
