Background

Acetylcholinesterase (AChE), also known as acetylhydrolase, is a crucial serine hydrolase that efficiently terminates signal transmission at neuromuscular junctions and cholinergic synapses by rapidly hydrolyzing acetylcholine (ACh) in the synaptic cleft. Beyond its fundamental role in neurotransmitter regulation, AChE has been found to participate in neuronal apoptosis and other cellular processes, suggesting a broader involvement in programmed cell death. In neurodegenerative diseases such as Alzheimer's disease, acetylcholine deficiency is closely associated with cognitive decline, making AChE an important therapeutic target. Currently, acetylcholinesterase inhibitors (AChEIs) are widely used in clinical practice for the targeted treatment of Alzheimer's disease and other forms of dementia.

Protocol

Experimental Method

Principle: Detect the ability of ACHE protein to cleave Acetylthiocholine.

Materials

  1. Assay buffer: 0.1 M Sodium Phosphate, 0.05% (w/v) Brij-35, pH 7.5

  2. ACHE/Acetylcholinesterase His Tag Protein, Human (SPA1701)
  3. Substrate: Acetylthiocholine (ATC) (A5626)

  4. 5’,5’-Dithiobis (2-nitrobenzoic acid) (DTNB) (D8130)

  5. 96 Well Clear Plate (701311)

  6. Plate reader (PerkinElmer, 405 nm)

Procedure

  1. Dilute ACHE protein to 0.02 μg/mL in assay buffer.

  2. Dilute substrate to 200 μM in assay buffer containing 100 μM DTNB.

  3. Add 50 μL of diluted ACHE protein to the clear plate. Include a control group with assay buffer instead of ACHE protein.

  4. Add 50 μL of substrate/DTNB mixture to initiate the reaction.

  5. Read the signal at 405 nm in kinetic mode for 10 minutes.

  6. Calculate specific activity:

Specific Activity (pmol/min/µg) =

Adjusted Vmax* (OD/min) x well volume (L) x 109 nmol/mol

ext. coeff** (M--1cm--1) x path corr.*** (cm)

x amount of enzyme (μg)

*Substrate blank correction

**Extinction coefficient 13260 M¹cm¹

***Path length correction 0.32 cm

Good for (non-destructive) small dogs
Color: Red
Bought for a small dog, would not recommend for a destructive chewer. After some initial hesitation this has become her favorite toy and keeps her entertained for a good while. The different settings provide some variety to how she can play with it. The battery only lasts about an hour, but it does charge quickly. As others have noted the string is garbage.

Great for small dogs with a big prey drive!
Color: Red
He loves it!! I have a small dog with a big prey drive and he goes absolutely crazy for this! A big plus for taking the boredom away, really perking up his mental attitude. It's also (at this moment) much cheaper than others that look very similar. Super easy to charge. My dog is a rescue and never learned to play with toys, has zero interest in stuffed toys and doesn't really understand play, but this isn't play to him: It's what he was born to do. He gives it a hearty Two Paws Up!

Our Yorkie Puppy Loves This Toy!!!
Color: Red
Warning: your pet may become completely obsessed I’ve had this toy for about a year for my Yorkshire Terrier, Gia, and it’s easily the most successful toy purchase I’ve ever made. We even had to replace the first one after she loved it a little too much (chewed the string off), and have since bought more as gifts for friends’ pets. Great toy for small dogs (too fragile for bigger dogs) and active, young cats.) What we love: * Endless engagement: Gia ignores all her other toys and goes straight for this every time * Interactive features: Subtle beeping + changing colored lights + unpredictable rolling patterns keep her hooked * Energy burner: Perfect for indoor play on hardwood floors—she will chase it nonstop * Rechargeable: Battery lasts several hours, even with frequent use * Durability: Holds up well considering how rough she plays Real-life note: Gia will literally bring this toy to me to get me involved—and I have to limit her playtime because she won’t stop on her own. Bottom line: If you have a high-energy dog (or even a playful cat), this isn’t just a toy—it becomes their favorite activity… and possibly their entire personality.

I wanted it to be a good fit
Color: Red
This toy garnered my dogs attention for a day. We tried it inside but there's too many places it would go under. We tried ot outside and it was powerful enough in the grass so tried concrete patio. My dog lost interest sadly. It wasn't a good product for my dog.
