| Usage | 1. Place the cell plate in the sterile operating table. 2. Add 10% volume of MTT solvent to each well of the cell plate. [Preparation of MTT solution: Dissolve 25mg of MTT with 5ml of MTT solvent to prepare a 5mg/ml MTT solution. It can be used after preparation, or stored directly at-20 ℃ in the dark from light, or stored at-20 ℃ in the dark from light after appropriately packaging as needed.] 3. Put the cell plate back into the incubator and continue to incubate for 3-4 hours. When doing comparative experiments, the incubation time should be consistent. 4. After incubation, take out the cell plate from the incubator and dissolve it according to the following method to produce MTT formazan crystals. (1) If it is an adherent cell, discard the culture medium. Add the same amount of Formazan Solvent as the initial volume. The volume of dissolved fluid may vary per well, but the total volume must be consistent for comparison. (2) If the removal of non-adherent cells or culture medium will cause the loss of MTT formazan, add the same amount of lysate as the initial volume directly to the culture medium. (3) Readings must be completed within 1 hour after addition of Formazan Solvent. 5. Gentle shaking in a shaker can enhance dissolution. In some cases, particularly when the cell density is high, repeated pipetting is required to completely dissolve the MTT formazan crystals. 6. Spectrophotometer to measure OD570 and reference wavelength OD690. 7. The 96-well plate can be detected on a microplate reader equipped with a suitable filter. 8. Other types of multi-well plates need to be spectrophotometrically detected with a cuvette or plate reader of appropriate size. |

| Description |
The MTT assay, also known as MTT colorimetry, is a method to detect cell survival and growth. The detection principle is that succinate dehydrogenase in mitochondria of living cells can reduce exogenous MTT to water-insoluble blue-violet crystalline Formazan and deposit it in cells, while dead cells do not have this function. Within a certain range of cell numbers, the amount of MTT crystals formed is directly proportional to the number of cells. The greater the absorbance, the stronger the cell activity, indicating that the drug toxicity is less. This method has been widely used in the activity detection of some bioactive factors, large-scale anti-tumor drug screening, cytotoxicity test and tumor radiosensitivity determination. | General Notes | 1. Since a 96-well plate is used for detection, if the cell culture time is long, we must pay attention to the problem of evaporation. Can't beat the price if you have smaller dogs

This is my 2nd one, which was bought for my mom's dogs. They absolutely love it when they come to my house. I live that they have to get their nose off the floor to figure it out and that there are more than 1 tube that they have to flip (think about). My only thumbs down isn't a big deal but would be the hard base. The treats fling off this base and can end up anywhere in the room. I think if it wasn't so hard, they wouldn't have to walk away from the activity or have to fight over who get the "reward" (It's usually the other dog waiting to chase where they fly off to). It doesn't flip over unless they run into it which doesn't happen often, although our dogs are all under 15 lbs. Imnsure if a bigger dog were to use this, it wouldn't stand up well and may break. It was easy to put together, and functions very well. Our dogs think this is the most fun toy out of all the interactive togs we have (which is A LOT).

Amazing!!!

This is an amazing mental stimulation toy for my dog! And it’s a perfect one for dogs who are learning. He picked it up right away and loves it! I can’t wait to get the other ones they have. Seriously so amazing. My very active Aussie can spend 30 minutes doing this and not get bored.

Great for bored dogs

Great dog puzzle! My pup loves this, he did chew the peg portion once he got them out so they can be put back in. They are tough for small dogs to get out so I think he plotted it out.

Actually Holds up

I must say I had low expectations when buying this bone based on prior experience of similar products. I'm happy to say I was pleasantly surprised. I bought this bone off Amazon in the beginning of December, I wanted to wait a while for my review to see if it would actually hold up. So far so good. There are several factors that set this bone above similar products.
First let me be clear that this is the Beef bone not the chicken etc. (I'm told there is a huge difference)
1. The first thing that impressed me is that my dog actually loves to chew on it. I know this may sound weird but I tried the puppy pack of nylabones and a bit larger one. He was interested in them but it didn't last and he wouldn't touch them anymore. This bone on the other hand is his absolute favorite. He carries it around everywhere. Even though it's beef flavored it doesn't have a noticeable smell or residue and it doesn't get slimly like a real bone.
2. This bone is extremely durable. I have a chew happy 10 month old lab puppy with razor teeth and majoring chewing skills. Trust me, my ruined shoes are proof :) It's been almost two months and I can just see where the bone has worn down. The down side to this is it is extremely heavy, not ideal to be leave around to step on, especially because of the way the bone breaks down. As they chew on it is almost peals away into tiny, tiny chunks. This is great compared to a kong stick that he had where he was swallowing large pieces of a rubber/plastic material. The downside is that as it "peals" it leaves the bone very sharp, with little spikes around where they chew. So far my dog doesn't seem to mind and it doesn't seem to make his mouth bleed or anything so that's a minor negative. Just don't stub your toe on it! Trust me!
Overall after trying multiple brand/types and flavors this is the only one that gets my seal of approval.

This is a great option for tough chewers

This is a great option for tough chewers, although if your dogs are anything like ours, there is a procedure to ANY new toy no matter how "tough". If I gave them this Nylabone and they went unsupervised day one, they would wreck it and it'd be in the trash within the hour. I say this to tell you, if you have a top 10 bite strength aggressive chewer, that you shouldn't expect a miracle.
If instead, I give it, take it away when they are working it too hard or being particularly destructive, and then give it back- and do this a few times for a week or so, then they'll chew it in a less aggressive fashion- they'll enjoy it and it lasts a long time. I don't know what it is about the first few days, but when something is "new" it has to be destroyed.
When it's been in the house a few days, they are rough but not as bad. Like the ones listed "virtually indestructible" will last 6 months as long as we have multiple around. It will show damage, there will be shrapnel in the carpet that will scratch up your feet, but it'll be worthy of play still. I consider 6 months in our house worthy of 5 stars.
This has been true for 2 Dobermans and a German Shepherd- and all of them have proven ability to destroy these products day 1 if I don't behave appropriately on introducing the new toy. If your dog wrecks these in the first 10 minutes, I'd say you probably need to work on supervising new toy play, and taking it away when they become to aggressive. This is a training issue, it can be done!
