Background
ICOS, a CD28 family receptor also designated as AILIM/CD278, functions as a crucial immunoregulator. It potently augments T-cell responses against foreign antigens through promoting proliferation, lymphokine secretion, and enhanced cell-cell interactions while facilitating B-cell antibody production. Although not affecting IL-2, ICOS induces IL-10 synthesis and sustains T-cell viability by preventing apoptosis in pre-activated T-cells. Its pivotal role in CD40-mediated immunoglobulin class switching underscores its importance in humoral immunity. Structurally, ICOS forms functionally active homodimers stabilized by disulfide bonds. Collectively, these properties establish ICOS as a master regulator coordinating T-cell activation, B-cell help, and antibody diversification in adaptive immunity.
